Thus, telomere length and telomerase activity are crucial for cancer initiation and the survival of tumors. Several pathways that regulate telomere length have been identified, and genome-scale studies have helped in mapping genes that are involved in telomere length control. Additionally, the nature of telomerase regulation is currently only poorly understood. More work on the tumor specific regulation of telomerase activity might provide either more opportunities for telomerase inhibition, or more skepticism, as a tumor cell might possess mechanisms for upregulating telomerase activity in the presence of inhibitors. Therefore, aging and cancer are two ends of the same spectrum. Inhibition of telomerase in cancer cells may be a viable target for anticancer therapeutics, whereas expression of telomerase in normal cells may extend healthy lifespan, especially for patients with inherited telomere spectrum disorders ( 104 ). 2021-04-14 · Telomerase is an important drug target for cancer.

Telomeres progressively shorten with age in somatic cells in culture and in vivo because DNA replication results the loss of sequence at the 5' ends of double-stranded DNA. Telomerase, a eukaryotic ribonucleoprotein (RNP) complex (26– 33), helps to stabilize telomere length in human stem cells, reproductive cells and cancer cells (35, 36) by adding TTAGGG repeats onto the telomeres using its intrinsic RNA as a template for reverse transcription .
In addition, normal human cells including stem cells have lower telomerase activity and generally maintain telomeres at longer lengths compared to cancer

This gain-of-function mutation upregulates TERT transcription . 2021-03-07 · Telomerase and Cancer Cancer cells multiply rapidly, which would normally result in shortened telomeres. Cancer cells make telomerase, however, preventing the telomeres from becoming so short that the cells can no longer survive.

Telomerase therapy is not only unlikely to result in an increased risk of cancer but is likely to lower the risk of cancer compared to age-matched patients not treated with telomerase therapy. Review of available data suggests that cancer should not be considered a significant risk to patients undergoing telomerase therapy, but the actual outcome will require human trials. Telomerase is an attractive target for diagnosis and therapy since it is expressed in over 90% of breast cancer cells, while it is not expressed in most normal cells. Whereas most of the progress on the role of telomerase in breast cancer has been in diagnostics, research into telomerase inhibitors is increasing.
